WHY DOES mRNA SEQUENCE DESIGN MATTER FOR IN VIVO CAR-T?
For an in vivo CAR-T program, the mRNA sequence is one of the levers that decides how much CAR protein T cells make, and how much gets made in cells the program didn't mean to reach. In mRNA-based in vivo CAR-T, the CAR is usually encoded in mRNA and delivered in lipid nanoparticles, so the patient's own T cells produce the receptor. Lipid nanoparticles often end up in the liver, so keeping expression low in off-target cells can matter as much as raising it in T cells.
nCode's goal is more protein per milligram of drug, made selectively in the target cell type. The same protein can be encoded by a very large number of mRNA sequences, because of codon choice and the untranslated regions (UTRs) around the coding sequence, and those choices can change how much protein a cell makes and how long the mRNA lasts.
nCode's early-access work already includes in vivo CAR-T, with an academic center working on hematology-oncology programs.
